Larter and husband Hayes MacArthur were married in 2009, after three years of dating.
When Larter was pregnant with their first baby, the couple went to Europe to try and keep the news of her pregnancy away from the media.
Their 3-year old son, Theodore, was born in 2010.
The 38-year old former star of the hit series, "Heroes", was glowing as she rubbed her baby bump and made the announcement on Fallon's show.
The talk show host brought up the fact that Larter was also pregnant when she guested on his show three years ago.
Larter laughed at the coincidence and joked, "Yeah, I came on and it was pregnant and it went so well. So I told my husband I was going on Fallon so I gotta get pregnant!"
Fallon, who is also enjoying life as a new father, bent down and spoke to the baby in Larter's belly, saying "Hey buddy!"
Fallon and his wife, producer Nancy Juvonen, have a daughter born via surrogate. The child, Winnie, was born last year.
Larter shared that she's been trying to conceal her baby bump, especially while shooting for the spy drama, "Legends". Her method for trying to divert attention from her growing belly was wearing blazers and push-up bras.
When asked by Fallon how she managed to keep the secret from other people, Larter joked, "I just keep telling [people] I've just been eating so many cheeseburgers."
The actress also shared details about cooking with her son, her new cookbook and "Legends".
"Legends", which also stars former "Game of Thrones" actor Ned Stark, will air every Wednesday on TNT.
